Investigation the elicitation of inflammatory responses by Resilon and guttaperchaon human monocyte cell line THP-1

Objective: In this in vitro study, the induction of inflammatory responses to the filler materials which usedtoday (Resilon and Gutta-percha) was investigated on the human monocyte cell line THP-1.Methods: Three millimeters of gutta-percha and Resilon were placed in direct contact with the THP-1 cellsuspension after disinfection in sodium hypochlorite solution on the ninety-six plates. Root filler materialswere evaluated for both LPS and non-LPS. The positive control group contained LPS, and the negativecontrol group contained only the culture medium containing the cell. Production of IL-1β and TNF-α afterexposure to the tested materials was evaluated by ELISA.Results: Production of IL-1β in all groups increased as time-dependent. Gutta-percha's ability to produceIL-1β was greater than Resilon. In both gutta-percha and Resilon groups, LPS contamination increasedTNF-α production.Conclusion: There was no statistically significant difference in Resilon and gutta-percha elicitsinflammatory cytokine production (either in terms of and gene expression or protein synthesis).
