Bioinformatics evaluation of hsa-miR-۱۳۹-۵p signaling pathways and its associated rs۵۶۲۰۲۴۱۹ function in patients with acute myeloid leukemia

Abstract:

Background and Aim: Acute Myeloid Leukemia (AML) is the most common acute leukemia in adults. The identification of recurrent genetic mutations, such as FLT۳-ITD, NMP۱ and AML۱, has helped refine individual prognosis and guide management. The translocation t(۸;۲۱), leading to a fusion between the RUNX۱ gene and the RUNX۱T۱ locus, was the first chromosomal translocation identified in AML.Methods: To get more information we used NCBI, miRNASNP, miRBase, miRWALK۲.۰, DAVID databases and KEGG pathway as the bioinformatics tools.Results: Investigations at the rs۵۶۲۰۲۴۱۹ locus indicated that an allele altered to G, which may effect on function of the miRNA associated with this region. Finally, hsa-miR-۱۳۹-۵p as one of the most important miRNAs related to this gene and given the confirmation of the role of hsa-miR-۱۳۹-۵p in AML through bioinformatics databases, this miRNA selected for this analysis.Conclusion: Studies on this rs۵۶۲۰۲۴۱۹ have shown that when the SNP locus is in the G allele, the miRNA binds to it less than the wild an allele. Due to the increase action of CSF۱R, GM-CSF, MPO, IL۳, so block of differentiation. We predicted that a person with G allele is more susceptible to AML by increasing the expression of the AML۱.
