Abstract:

Background and Aim: Diabetes mellitus comprises of a series of metabolic disorders featured by abnormally high glucose level in blood. Type ۲ diabetes mellitus (T۲DM) develops because of insulin resistance of human organs and insufficient secretion of insulin from pancreaticβ-cells. The lncRNA growth-arrest specific transcript ۵ (GAS۵) is a ۵′-terminal oligopyrimidine class of genes which regulates cell growth, proliferation and survival. GAS۵ acts as a riborepressor by repressing transcription of glucocorticoid receptor. It has been shown that inhibition of mTOR can increase the level of GAS۵. In addition, it interferes with carbohydrate metabolism and increases likelihood of developing diabetes. However, the exact role of GAS۵ in diabetes is not fully understood. The aim of this study was to investigate the expression level of GAS۵ in patients with T۲DM.Methods: A total of ۴۰ genetically unrelated subjects including ۲۰ healthy controls and ۲۰ patients with T۲DM participated in this study. Total RNA was isolated from blood samples, using TRIzol according to the manufacturer's protocol. Synthesis of cDNA was performed. Primers for real time PCR were designed, synthesized and specificity of the binding of primers was confirmed by gel electrophoresis. In this study the GAPDH gene was used as an internal control gene. The LncRNA GAS۵ expression level was evaluated by real-time PCR and the results were analyzed by SPSS, PRISM.Results: We observe a marked decrease in GAS۵ expression in diabetic samples compared to non-diabetic (P<۰.۰۵).Conclusion: In conclusion, this is a first report of GAS۵ expression in T۲DM. More research is needed to fully elucidate the underlying molecular mechanisms of T۲DM susceptibility.
