Abstract:

Background and Aim : Leptospirosis is a zoonosis caused by infection with pathogenic strains of the bacterium Leptospira . This infection has a worldwide distribution, and is an emerging/re-emerging public-health problem, particularly in large urban centres of tropical and subtropical regions. Uveitis represents the ۵th leading cause of visual loss in Europe with a prevalence of approximately ۳۵–۸۰ per ۱۰۰,۰۰۰۱ causing approximately ۵%–۲۰% of blindness. Interleukin (IL) ۱۷ is a proinflammatory cytokine released by helper T cell-۱۷ (T-helper ۱۷) already known to play a defense role against microbes. Recently, the association of Th-۱۷ cells or IL-۱۷ with ocular inflammatory diseases such as uveitis, scleritis and dry eye syndrome was discovered. In this study, researchers aimed to investigate role of IL-۱۷ in inflammation of eye by leptospirosis.Methods : Sample of eye (aqueous humor) collected from ۵۰ mice fall into two groups (healthy control and patient) for RNA extraction. The extracted mRNA was subjected to cDNA synthesis and the resultant cDNA was used for analysis of IL-۱۷ using real time PCR. The IL-۱۷ was also measured by ELISA (enzyme-linked immune-sorbent-assay).Results : Results of the present investigation have indicated that all of leptospirosis patient exhibited elevated levels of IL-۱۷ when compared to control. The mean level of IL-۱۷ was ۱۳۶.۳۸±۸ pg/mL and ۲۱.۰۷±۲pg/mL in leptospirosis patients and controls, respectively.Conclusion : According to the findings of the present study the IL-۱۷ axes may play fundamental parts in pathogenesis of leptospirosis by recruitment of immune cells toward eye inflammation. These axes may possibly serve as therapeutic factors for therapy of leptospirosis. The demonstration of IL-۱۷ involvement in various ocular and infectious disease is supporting evidence for recent reports (۱۳, ۱۹) showing that Th-۱۷ cells are important in the pathogenesis of ocular surface diseases. We demonstrated in this study that elevated IL-۱۷ levels.
