Abstract:

In this research, we attempted to evaluate different crude antigens obtained from mycobacterial species using an indirect ELISA test. The purpose of this assessment is to detect the infection of different mycobacteria in infected livestock.Methods : Five different strains of Mycobacteria (M. tuberculosis, M. phlei, M. bovis, M. avium subsp. Paratuberculosis, and M. bovis AN۵) were cultured and precipitated by TCA ۴%. The SDS-PAGE of crude antigens of different mycobacteria was accomplished. Protein concentrations determined with Lowry protein assay. After preparing the crude antigens of mycobacteria, the ELISA test was performed, the results of the ELISA test were compared with the PPD skin test and data analysed with SPSS software.Results : Study showed, all five strains had more than ۹۲% detection ability in healthy animals. The highest sensitivity of ELISA tests to M. bovis AN۵ antigen is greater than ۸۳% and the highest diagnostic specificity of M. avium subsp. Paratuberculosis is ۹۵.۸۳% and the highest efficiency of diagnostic tests is over ۸۳% concerning this antigen.Conclusion : In conclusion, this study concludes that M. avium subsp. Paratuberculosis and M. bovis AN۵ antigens are suitable candidates for the design of diagnostic ELISA due to their sensitivity, specificity, and efficiency.
