Short communication: Effect of light and nitrogen concentration on the growth and lipid content of marine diatom, Chaetoceros calcitrans

Abstract:
The effects of light intensity (µ mol m-۲ s-۱) and different levels of Nitrogen was made on the biomass and lipid of microalgae Chaetoceros calcitrans under laboratory conditions in the north of Iran in ۲۰۱۶. The pure strand of C. calcitrans with F/۲ media (۸ sets of ۳ flasks) were placed in front of the light source at values of ۸۰, ۱۶۰, and ۲۴۰ µ mol m-۲ s-۱. The cell count was collected in lasted ۷ days. Every day cells were counted recorded. Maximum cell densities were approximately ۲۷۹, ۲۹۹, ۳۹۵ and ۴۰۲×۱۰۶ cells ml−۱in ۲۵, ۵۰, ۷۵ and ۱۰۰ % nutrient levels respectively, up to the ۲۰th day of media enrichment, also maximum cell density was ۹۸×۱۰۶ cells ml−۱ in ۱۰۰% nutrient level and ۲۴۰ µ mol m-۲ s-۱ up to the ۲۰th day. Results showed that the lipid yield decreased significantly with increasing light levels. The total lipid was significantly higher (۱۴.۴۱%) in control compare with ۲۵, ۵۰, ۷۵ and ۱۰۰% nutrient levels. The total lipid at ۸۰, ۱۶۰, and ۲۴۰ µ mol m-۲ s-۱ were ۱۳.۳۱, ۱۲.۰۹ and ۱۰.۱۷% respectively. The production and storage of lipids by microalgae in response to variations in environmental factors are specific for each microalgae species.
