Scrotal hyperthermia is one of the most important causes of infertility in males. Exposure to iron superoxide nanoparticles significantly reduced sperm concentration, motility and viability. However, co-administration of Fe2O3 nanoparticles with CoQ10 and curcumin greatly improved sperm parameters.Background: Scrotal hyperthermia is one of the most important causes of infertility in males and this increase in testicular temperature is due to lifestyle, work habits and environmental conditions. In the present study, the antioxidant effects of coenzyme Q10 and curcumin along with iron superoxide nanoparticles on sperm parameters of rats with testicular hyperthermia were investigated.Materials and Methods: In this study, 48 rats were divided into two groups: normal and scrotal hyperthermia. Both groups were given iron superoxide nanoparticles subcutaneously for 10 days. Then testes and epididymal tissue were eliminated and after evaluating sperm parameters, parameters such as viability, motility, concentration and morphology of sperm were examined. One-Way ANOVA was used for data analysis.Results: Exposure to iron superoxide (Fe2O3) nanoparticles significantly reduced sperm concentration, motility and viability, as well as increased proapoptotic BAX gene and decreased expression of BCL2 anti-apoptotic gene. However, during co-administration of Fe2O3 nanoparticles with CoQ10 and curcumin greatly improved sperm parameters in hyperthermia rats.Conclusion: Exposure to Fe2O3 nanoparticles reduces sperm parameters and increases apoptosis also has a negative effect on fertility. The results showed that the positive effect of curcumin and CoQ10 antioxidants along with Fe2O3 nanoparticles has a protective effect against spermatogenic cell death caused by hyperthermia. Therefore, the synthesis of green nanoparticles by prescribing antioxidants such as CoQ10 and curcumin is recommended for the treatment of hyperthermia.
