Abstract:

A magnetic molecularly imprinted polymer (MMIP) was synthesized and evaluated for selective extraction of sufentanil from human plasma and urine samples. The synthesis of the magnetic nanoparticles were performed by chemical coprecipitation method then were coated with silica layer and vinyl moieties. MIP layer was synthesized on the surface of modified magnetic nanoparticles in the presence of sufentanil (SUF) as template, methacrylic acid (MAA) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as the cross-linker monomer, ۲,۲′-azobisisobutyronitrile (AIBN) as the initiator. The synthesized MMIP and MNIP were characterized by FT-IR spectroscopy, UV-VIS spectrophotometry and scanning electron microscope (SEM). Extraction of sufentanil molecules from modeling solutions and real samples is followed by two steps: sorption and desorption. In this study, solid phase extraction using the magnetic imprinted polymer has been optimized with the experimental design approach for sufentanil optimum determination and factors that probably influence the extraction process, such as pH of loading sample, eluent solvent, volume of the eluent solvent and sorbent mass, were evaluated to achieve the highest extraction efficiency. Batch rebinding capacity of sufentanil was determined from the derived. The recovery percent, limit of detection (LOD), limit of quantification (LOQ) and relative standard deviation (RSD %) for biological samples were ۹۵% , ۱.۲ μg l-۱ , ۵ μg l-۱ and ۴% by introduced selective solid phase extraction respectively. Furthermore, the imprinted polymers were successfully applied as a specific solid-phase extractants combined with high performance liquid chromatography (HPLC) for determination of sufentanil in the biological samples.
