Gene expression analysis of Parvovirus B۱۹ in primary and continuous cell lines

Abstract:

Backgrounds: Parvovirus B۱۹ is a widespread pathogenic virus that only causes infection inhumans. The virus can complete and multiply its lytic cycle when it infects its host cells. Themain host of the virus is erythroid progenitor cells in the human bone marrow. Studies haveshown that the virus has the ability to enter some cells but cannot multiply in them. In this study,expression of non-structural (NS۱) and capsid (VP۱ and VP۲) viral genes in different cell lineswere evaluated.Materials and Methods: Gene expression analysis was performed in several primary(mesenchymal stem cells and cord blood cells) and continuous cell lines (HEK-۲۹۳, MCF-۷,K۵۶۲). Total RNA from the cells was extracted, cDNA synthesized and reverse transcriptionPCR was performed using Corbett PCR machine by specific primer designed against NS۱, VP۱and VP۲ genes.Results: The results demonstrate high expression of NS۱ gene in selected continuous cell lines.Also, detectable expression of NS۱ gene was observed in selected primary cells. On the otherhand, VP۱ and VP۲ were not expressed in any of the studied cell lines.Conclusion: Expression of viral genes in several cells indicates strong infective capability ofB۱۹ virus. Especially in cord blood cells, it shows that the virus can infect fetal cells eitherduring pregnancy or immediately after birth. Also, presence of viral gene expression inmesenchymal stem cells can be a remarkable subject which should be considered in safety ofstem cell products.
