Association between rs۹۳۴۴ polymorphism of cyclin D۱ gene (CCND۱) on breast cancer in Iran

Abstract:
Background: Cell cycle regulator cyclin D۱ (CCND۱) is a pivotal regulator for G۱/S phasetransition, a critical part of carcinogenesis initiation. Breast cancer comprises a veryheterogeneous group of cancer cells, but little is known about what is wrong in the genome ofthese patients. This study investigated contribution of CCND۱ (rs۹۳۴۴) in the breast cancersusceptibility.Materials and Methods: Blood samples were collected from ۵۰ patients diagnosed with Breastcancer and ۵۰ people as controls. Genomic DNA was extracted from whole-blood samples usingTriton-X۱۰۰ and genotyped by tetra-primer amplification refractory mutation system PCR (TARMS-PCR). Specific oligonucleotide primers for amplification of the CCDN۱ were designedby Oligo۷ software. Finally, the statistical analysis was performed by MedCalc software.Results: The results of this study showed a significant difference in genotype distribution ofrs۹۳۴۴ CCDN۱ polymorphism between breast cancer patients and controls (p=۰.۰۳).we indicatedGG genotype frequency in breast cancer patients is significantly higher than controls.Conclusion: Our study suggests that the polymorphism rs۹۳۴۴ in cyclin D۱ gene has associationwith cancer risk in Guilan population, indicating that CCND۱ rs۹۳۴۴ may be an importantbiomarker to prediction of breast cancer risk. However, future studies with more patients andcontrols are needed to confirm the results. Also, additional evidences are necessary to furtherclarify the findings of this research.
