Effect of Memantine on Expression of NAT-Rad۱۸, Rad۱۸ and Sorl۱ Genes in Rat Model of Alzheimer's Disease

Abstract:
Background and Objective: Dysregulation of long-term expression of non-coding RNAs (lncRNAs) has a potential role in progressive brain disorders such as Alzheimer's disease. This study aimed to analyze the apoptosis and expression of ۵۱A and NAT-Rad۱۸ lncRNAs and their target genes in brain tissue and peripheral blood mononuclear cells (PBMCs) of the rat model of AD, before and after memantine treatment.
Materials and Methods: Twenty-eight male Wistar rats were divided into four groups: ۱. Normal control (n = ۴), ۲. Sham-operated (n = ۴), ۳. Alzheimer's control (n = ۱۰), and ۴. The experimental group (n = ۱۰) was treated with memantine. The qPCR and TUNEL tests were used to detect the lncRNAs expression and apoptosis.
Results: Sorl۱ gene was reduced in brain tissue of Alzheimer’s control (p = ۰.۰۱۶) and PBMCs of Alzheimer's control and experimental groups (p = ۰.۰۰۲ and p = ۰.۰۰۱ respectively). The expression of NAT-Rad۱۸ and Rad۱۸ genes increased in brain tissue of Alzheimer's control group (p = ۰.۰۰۲ and p = ۰.۰۴ respectively) while reduced in PBMCs of Alzheimer's control and experimental groups (p = ۰.۰۰۵ and p = ۰.۰۴۵ for NAT-Rad۱۸, p = ۰.۰۱ and p = ۰.۰۰۶ for Rad۱۸).
Conclusion: ROC curve analysis showed ۱۰۰% sensitivity and ۸۵.۷% specificity for the Sorl۱ gene with ۰.۹۱۱ under the curve area and ۱۰۰% sensitivity and ۱۰۰% specificity for NAT-Rad۱۸ and Rad۱۸, separately with one under the curve area. Decreased expression in Sorl۱, NAT-Rad۱۸, and Rad۱۸ genes can be used as blood biomarkers for diagnosis independently. However, studies on Alzheimer's patients are needed.
