Blending of Polyethylene Glycols with Different Molecular Weights:Physical Properties and Ibuprofen Delivery abstract

Polyethylene glycols (PEG) in different molecular weights are used as water-soluble carriers todeliver a long list of therapeutic agents to rectal mucosa. However, the necessary physical propertiesor drug delivery kinetics are rarely achieved using a single PEG with a definite average molecularweight and polydispersity index implying a certain need for blending. To this end, PEGs in differentmolecular weights (MW=2000-10000 g.mol-1) were used in alone or as 50:50 dual blends toformulate ibuprofen (IBU) suppositories. Physical and drug delivery characteristics of the deviceswere investigated supporting that 50:50 blends of PEG 2000:10000 and PEG 4000:10000 presentedthe optimal physical properties and IBU release kinetics. According to the results, difference inmolecular weight of PEGs affect drug release kinetics only at early times, i.e. 20 minutes
