Optimization Of Bacteria Producing Asparaginase Enzyme In Hard Layer Based On Response Surface Method abstract
Asparaginase is an enzyme that is used in the food industry as well as medicine. Its medicinal uses are the treatment of acute lymphocytic leukemia, acute bone marrow leukemia, and non-Hodgkin's lymphoma, and it can be administered by subcutaneous, intramuscular, and intravenous injection Without it, cancer cannot make DNA. The preliminary investigation using the Plackett-Burman design showed that among the tested carbon sources including: rice bran, wheat bran, rice stalk, wheat stalk, rice bran has the best effect in increasing enzyme production and among the tested nitrogen sources including: asparagine , yeast extract, peptone, tryptone, asparagine showed the best effect in increasing asparaginase production in solid medium (p<0.05). In the next stage of optimization, by examining 4 factors of rice bran, asparagine, incubation temperature, incubation time at 5 levels on the production of asparaginase by using the RSM method, the amount of enzyme production was 24.69 U/g. In the solid substrate, the amount of enzyme production was 3 times compared to using the experimental design using the Plackett-Burman and RSM methods. Also, research was conducted to determine the characteristics of the asparaginase enzyme produced by the isolated bacterial strain. The maximum amount of experimental enzyme production obtained in the solid bed was at the concentration of rice bran 1.5% w/w, the concentration of asparagine 0.5% w/w, temperature 30°C, and duration 72 hours. The results indicate significance. The production of asparaginase in solid substrate using rice bran waste substrate is due to its cheap price and abundant availability and high potential in enzyme production.
