Evaluation the effect of combination injection of rat oral mucosal stem cells and platelet rich plasma and vitamin C on rat Achilles tendinitis

Background: Tendon is a type of connective tissue that connects muscle to bone. Healing of tendon injuries is associated with scar tissue without elasticity. Old medical and surgical treatments often leave a lot of granulation tissue and scar tissue. Stem cells exert their healing effect by regulating the inflammatory phase. Vitamin C plays a vital role in hydroxylating proline, which is the precursor of collagen synthetase, and by reducing oxidative stress, it leads to increased survival of implanted cells.Materials and methods: Eighteen male Wistar rats were randomly divided into negative control and two treated groups. Tendonitis was induced by collagenase in Achilles tendons. After two days, a single dose of the PRP was injected into the injured tendons and vitamin C injected intramuscular. Six rats received single dose of PRP and stem cells intratendonal and vitamin C intramuscular. In the control group, animals were not treated with anything. Following 7 and 14 days, the tendons were dissected out and the samples were used to perform histopathological examinations.Results: Both treatment groups showed better repair compared to the control group. Hematoxylin-Eosin histopathological evaluation showed that in the stem cell recipient group, collagen strands have a better arrangement.Conclusion: PRP and vitamin C lead to improved tendon repair and better arrangement of collagen fibers.Mesenchymal stem cells promote the repairing effect of PRP and vitamin C in tendinitis.
