Abstract:

The interaction between a Cu(II) complex containing gabapentin drug and ct-DNA was studied by emission, absorption, fluorescence techniques and molecular modeling. In fluorometric studies, the binding mode of the complex with ct-DNA was investigated using Hoechst as a fluorescence probe. Stern-Volmer quenching constants were measured at different temperatures which indicated static mechanism. The binding constant (K) earned from fluorescence quenching studies, which enhanced with temperature rising. Enthalpy and entropy changes during the interaction of complex with DNA were obtained by van't Hoff plot, which showed an endotermic process and complex mainly bound to ct-DNA by the hydrophobic attraction and the mode of binding is groove. AG value revealed a spontaneous reaction between DNA and complex. Optimized docked model of DNA-complex mixture confirmed the experimental results. In comparison with the previous study of the DNA interaction with gabapentin, these results show that copper complex has greater affinity to ct-DNA.
