Abstract:
With almost 9 million new cases each year tuberculosisremain of the most feared disease on the planet. Theannual incidence of tuberculosis ( TB ) among indigenousirainians stands at 14 cases per 100000 inhabitants.Golestan province after Sistan and baloochestan has thehighest number of TB patient in iran.in this study RFLPteqnique by pvu II enzyme and PGRS and DR probewas done. Two Sputum speciement were collected fromsmear positive patient in Golestan province. Sampleswere processed and culture on Lowenstein Jensen (LJ)slant for three weeks. DNA was extracted from twosample based on van embden protocol.first PCR assay targetting 16S rRNA genes was used foridentification of Mycobacterium spp. whole DNA RFLPwith Pvu II restriction enzyme and hybridized with DIGlabeled PGRS and DR probe. The results obtained byclonialMorphology, PCR showed that both isolates werebelonging to M.tuberclosis ,Genotyping of the isolatesby RFLP technique displayed that two isolates werebelonging to different strains of M.tuberclosis
