Abstract:

Prostate cancer is a major leading cause of cancer related death in men. Biebersteinia(Geraniaceae) has been widely used in herbal medicine of many countries including Iran. Inthis study we exposed DU 145, PC3 human prostate cancer cell lines, and HEK293 cells toethanol extract of B. multifida to evaluate the apoptogenic and cytotoxic activity of the plant.B. multifida was collected from Pivejan, Mashhad, northeast of Iran. Cells were cultured inRPMI-1640 medium and humidified air with 5% CO2. AlamarBlue® assay was used toindicate the cytotoxic activity of B. multifidi. Finally, flow cytometry was used to detectapoptotic cells. Viability of cells was inhibited by B. multifida dose-dependently. DU 145 cellswere more resistant than PC3 and HEK293 cells to the cytotoxic activity of B. multifida. Incomparison, , there was an increment in sub-G1 peak of cells exposed to ethanol extract of B.multifidi in flow cytometry histograms of untreated control and treated cells which shows theapoptosis induction with B. multifidi. Further analytical and mechanistic studies aresuggested on B. multifidi according to observed apoptogenic and cytotoxic activity.
