Abstract:

Early detection of Amyloid deposit could be effective in the diagnosis and Treatment of disorders such as Alzheimer’s disease (AD), Parkinson’s disease (PD) And systemic amyloidosis. In this study, we tried to synthesize the Benzofuranone Derivatives such as 6-Hydroxy-2-(3ˊ,4ˊ,5ˊ- trihydroxy benzylidene) benzofuran-3(2H)-one compound as anAmyloid detection probe to compare with a standard probe called Thioflavin T. β-Lactogluboline and Globin as a protein model were purified via Fractionation Method, and their quality were evaluated by SDS-PAGE and Bradford protein Assay. The Spectrophotometric analysis of the 3ˊ,4ˊ,5ˊ trihydroxy benzofuran compound such as UV-visible and fluorescence spectroscopy were considered on its individual and Special wavelengths. This synthetic compound was excited and the emission Spectra were recorded immediately in its own exclusive wavelengths while it were bound to the fibrils in comparison with Native/Amorphous aggregation. The results 3ˊ,4ˊ,5ˊ trihydroxy benzofuran compound was obtained in three different conditions, Native, Amorphous aggregation and Amyloid. We show that the 3ˊ,4ˊ,5ˊ trihydroxy benzofuran compound could selectively and specifically bind to Amyloid fibrils, apparently so much more than the ThT probe. Furthermore, our synthetic compound due to neutral charge and High Lipophilicity essence might cross the blood-brain barrier as an effective Probe. According to our result, the synthetic compound could be accounted as a Remarkable probe to detect in vitro β-amyloid plaques, but it should be Investigated further as potential probes for detecting β-amyloid plaques in the AD Brain.
