Abstract:

Nanoparticles have a potential role in a range of fields including medicine, technology, medical,industrial and etc. The use of fungi, plants and other environmentally friendly compounds for thesynthesis of nanoparticles is of particular importance due to the elimination of hazardouschemicals in the process. Due to the widespread use of the new nanoparticles with environmentallyfriendly methods (NPS) such as zinc oxide (ZnO) extensive research has been done inpharmaceutical and health fields. In this study, the cytotoxicity and anti-inflammatory potential ofzinc oxide nanoparticles were evaluated against MCF7 cells (Human breast cancer cells). The Mcf7cells were cultured in vitro to achieve the logarithmic growth phase then seeded and treated withvarious concentrations of NPs (0, 200, 400, 600, 800, 1000 μg/ml) followed by colorimetric cellsurvival assay using MTT. Morphological changes in the cells was determined using invertedmicroscope. The Real Time PCR assay was carried out on the synthesized cDNA to evaluate theexpression of genes (IL-1β, IL-10) involved in the immune system. ZnO NPs demonstrated moderatecytotoxic activity against Mcf7 cells with IC50 about 600 μg/ml and exhibited anti-inflammatoryeffect through dose-dependently suppression of IL-1β and increased IL-10 expression. Theexpression of IL-1β gene was reduced from 1 in untreated cells to 0.04 in treated cells with 800μg/ml of ZnO-NPs while the expression of IL-10 showed an increase of 1 to 7.2 at similarconcentrations.This study elucidates moderate toxicity effect of nanoparticles as well as excellentits anti-inflammatory potential in MCF7 cells and Provides grounds for the application ofnanomaterial s in therapies for a variety of diseases, including cancer, autoimmune and otherdiseases.
