Abstract:

Today, due to the exposure of human to deleterious materials some of the deadly diseases, including cancer, have become widespread among many types of cancer, most common type of liver cancer ishepatocellular carcinoma, in which chemotherapy with the use of drugs apply to kill or inhibit the growth of cancer cells. Schiff base is known to have biological activities such as antibacterial,antifungal, antitumor and antioxidant properties. The purpose of this study is to investigate the cytotoxicity effects of Schiff base on Hep G2 liver cancer cell lines. The MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay is cell viability test based on effect of material and exogenous material on alive cells. This test has done on 96-well plate. First cells were seeded on 96well-plate and incubated for 24 hours. Then, cells were treated with different concentration of Schiff base (200, 400, 600 and 1000 μg/ml) for 24, 48 and 72 hours. After that medium has removed andMTT added to each well and incubated for 4 hours. Then, DMSO was added in order to solve the dye. Plates were put in ELIZA reader and optical absorbance were read at 570 nm. The results obtainedhere, shown that the toxicity of Schiff base is dependent on concentration. Results showed that Schiff base, is able to inhibit Hep G2 hepatocellular carcinoma cells and values of IC50 at 24, 48 and 72 hours were 500, 480 and 470 μg/ml, respectively. our findings indicated that N,N′-dipyridoxyl(O- phenylenediamine) Schiff base is able to inhibit liver cancer cells, thus, this Schiff base probably couldbe used in suppression of cancer cells and cancer treatment
