Investigation of the effects of zn o nanoparticles synthesized via green chemistry in ferula assafoetida extract on breast cancer cell line

Abstract:

Cancer is one of the leading causes of death. Among the many types of cancer, the most common cancer which affects women is breast cancer. The breast cancer begins in the cells of the lobules, whichhave glands that producing and secreting milk. The use of metal oxides as anticancer agents has the advantage of improving cancer treatment. Zinc oxide (Zn O) nanoparticles are among the substancesthat are effective in treating cancer. The aim of this study is to synthesis Zn O nanoparticle and to evaluate its cytotoxicity on MDA cell line with MTT assay. The zinc oxide nanoparticles synthesizedusing a plant Ferula assafoetida. this plant is used to control the blood pressure and pain relief. The MDA cells were cultured in a laboratory and treated with Zn O nanoparticles. MDA-MB-231 cellswere seeded in 96-well plate. After 24 hours, cells were treated with Zn O nanoparticles for 24, 48 and 72 hours. At each of these times, MTT was added to each well and removed after 4 hours incubation.then 200μl DMSO added to dissolve insoluble crystals. Plates were read using ELIZA reader at 570 nm. The results showed that Zn O nanoparticles have increased cytotoxicity on the MDA cells, whichdepend on time and concentration. The result of MTT assay showed that IC50 values were 60, 40 and 31.2 μg/ml for 24, 48 and 72 hour-time intervals respectively. The percentage of cell viability decreasedwith increasing time and concentration. The percentage of viability increased from %84/46 to %11/83 in time of 24 to 48 hour time intervals.Findings indicated that Zn O Nanoparticles synthesized inFerula assafoetida extract have inhibited the growth of breast cancer cells
