Abstract:

Objectives and Background: Schiff bases as chelate ligands have played a key role in the chemistry of intermediate metals as well as in the primary group metals. Schiff bases are aldehyde- or ketone-likecompounds in which the carbonyl group is replaced with an imine or azomethine group. These ligands can easily create sustainability complexes with most metal ion intermediates. The metal complexesinteracting with the oxygen and nitrogen. Studies show that Schiff bases are effective compounds for inhibiting cancer cells. Recently a great deal of attention has paid to the therapeutic and therapeutic effects of these compounds; the most important of which are the anti-cancer effects and anti-oxidant properties. The aim of this study is to evaluate cytotoxicity of Schiff base (Manganese II complex fromN- N-dipyridoxyle) on MCF7 breast cancer cell line. Materials and Methods: MCF7 cell line was seeded in 96-well plates. cells cultured in RPMI1640 medium supplemented with 10% serum fetal bovine and1% penicillin /streptomycin. The cells were exposed to different concentrations of Schiff base (250, 125, 62.5, 31.2, 15.6 μg/ml) for 24, 48 and 72 hour-time intervals. After the incubation period, thetreatment medium was removed and MTT solutions were added to each well. After 4 h of incubation in the dark, the MTT was removed and DMSO was added to each well. Finally, the absorbance wasrecorded with ELISA reader at 570 nm. Results: results shown cytotoxicity of complex Mn+2 of N,N di pirodexil 1,4 butadiene Schiff bases on MCF7 cells. Schiff base is able to inhibit cancer cells withIC50 values of 500, 250 and 62 μg/ml at 24, 48 and 72 hour-time intervals, respectively. Conclusion: Results of this study indicated the inhibition effects of complex Mn+2 of N,N di pirodexil 1,4butadiene Schiff bases in the growth of MCF7 breast cancer cell line. Therefore, it may be possible to use this Schiff bases for cancer therapy. Keyword: Cancer, Schiff bases, MTT Assay, MCF7 cell line
