Abstract:

Colorectal cancer (CRC) is the third leading cancer in the world. Despite conventional therapy methods such as surgery, radiotherapy, and chemotherapy, the mortality rate of this cancer is very high. Oneobstacle to success with chemotherapy treatment is drug resistance and the weak response to treatment. Thus, there is an urgent need to develop novel anticancer agents that either exert a better efficacy than conventional therapy or improve them in the context of combinatorial regimens. Curcumin is a hydrophobic polyphenol that derived from turmeric rhizome (Curcuma longa).Anticancer activity of curcumin has been proven in various types of tumors in clinical and para-clinical studies. This study investigates the antitumor effects of a novel form of turmeric oleoresin in colorectalcancer cells. CT-26 cells were cultured in F12 medium with 10% FBS and 1% penicillin and streptomycin, then incubated at 37˚C and 5% CO2. In order to investigate the effect of cytotoxicity of turmeric oleoresin alone and in combination with 5-FU, the MTT assay was used. The level of cyclin D1 expression involved in cell cycle regulation was determined with quantitative RT-PCR. Turmeric oleoresin inhibits cell growth by regulating the Wnt/β-catenin pathway and decreasing the expression of the cyclinD1 gene expression. The combined treatment of CT-26 cancer cells with turmeric oleoresin and 5-FU decreases the dosage of the drug (IC50). This research has proved the anti-cancer activity of turmeric oleoresin in CT-26 colon cancer cell line alone and in combination with 5-FU and supports further studies on this potential therapeutic agent in the treatment of colorectal cancer
