Abstract:

Breast cancer is one of the leading causes of death in womenaround the world with a very high degree of mortality and morbidity.Conventional treatments use cytotoxic drugs which have high levels of sideeffects, affecting the patient’s quality of life. Therefore today’s pharmacology islooking into treatments with low side effects and maximum efficiency. TheHelicobacter pylori neutrophil-activating protein (HP-NAP) is a virulence factorthat attracts and activates neutrophils, and promotes their endothelial adhesionand the production of oxygen radicals and chemokines. In this study, we willevaluate, Interaction of recombinant HP-NAP Helicobacter pylori in MCF7 cellline.Methods and Materials: HP-NAP Protein expression was induced by adding1mM isopropyl-β-D-thiogalactoside to cultures of E. coli strain BL21transformed with pET- HP-NAP. Protein expression assessed with SDS-PAGEanalysis. MCF7 cell line was cultured with different concentrations ofrecombinant HP-NAP and cytotoxicity were evaluated by MTT assay.Results: SDS-PAGE analysis showed the expression of an approximately 22Dalton protein. MTT assay showed that recombinant HP-NAP in 10 and 20μg/ml concentration significantly inhibit tumor cells growth (P<0.001).Conclusion: This study is interventional study and results showed thatrecombinant HP-NAP by having immunomodulatory properties could be usedin several inflammatory and immunologic disorders. HP-NAP may be a newtool for future therapeutic strategies aimed in cancer immunotherapy.
