Abstract:

Breast cancer is the most common cancer in women worldwide and is aheterogeneous disease with several environmental factors and genetic changessuch as polymorphisms. Arg/Pro polymorphism at codon 72 of the P53 as oneof the most common site for genetic alteration in human cancers and ABCC1promoter polymorphism as a risk factor for progressive cancer has beeninvestigated in different populations. In this study, effect of two polymorphismsin 25 tissue samples from women with breast carcinoma and 30 control sampleswere analyzed by PCR-RFLP with BstUI and HpaII and PCR-SSCP. Thedistribution of CC, GC and GG genotypes of codon 72 of p53 were 8.00%,56.00% and 36.00% respectively in patients with cancer and 13.33% CC,56.67% CG and 30.00% GG in controls. The relative allele frequencies ofABCC1 promoter polymorphism were 54.00% A and 46.00% T in patients and36.66% for A and 63.34% for T in normal controls. With c2 test there isstatistical difference in the distribution of p53 codon 72 and G1666A of ABCC1polymorphisms between patients and control groups (p<0.05). Study of thesesamples with PCR-SSCP did not display any new band template. Our resultsdemonstrated that p53 codon 72 and G-1666A of ABCC1 polymorphisms mighthave a probable role in breast carcinogenesis in range of high population.
