One Step Plantlet Regeneration from Leaf Explants of Begonia semperflorens abstract
The capacity to obtain somatic organogenesis is very important tool for in vitro studies of plant species, and frequently a bottle-neck for successive steps in plant genetic transformation. We report here an efficient protocol for shoot bud differentiation from mature leaves of Begonia x semperflorens-cultorum. For this purpose, we tested 16 different combinations of NAA (1, 2, 10 and 20 µM) + BA (1, 2, 5 and 10 µM) and two light conditions (16h at 50 μmol m-2 s-1 and darkness). The combination MS + 1 µM BA + 1 µM NAA in dark conditions showed the best response concerning adventitious bud induction and plant regeneration from leaf explants (70% regeneration).
