Combination effect of CUR/SOR on MEK,ERK/ NF-κB in HepG2 cell line abstract
Introduction: Hepatocellular carcinoma (HCC) is the fifth most common malignancy and the third major cause of cancer-related mortality worldwide. During the past decades, the incidence of HCC has increased. Sorafenib (SOR) is the first oral multi-kinase inhibitor which decrease cell survival rate with advanced HCC. Curcumin (CUR) has several pharmacological effects, including anti-oxidant, anti-inflammatory, anti-tumour. The aim of this study was to investigate the in vitro effects of SOR and CUR and their combination on proliferation and survival of human HCC cell line. Methods: The effects of SOR and CUR on the growth and viability of HepG2 cells were assayed by MTT method. The HepG2 apoptosis and cell cycle distribution were detected by flow cytometry. The altered expression of genes (MEK1,MEK2,ERK,Cyclin D1,NF-κB, PTEN) was confirmed by quantitative Real Time PCR.Results: Effective dose and IC50 of SOR and CUR in HepG2 cell line is 5μM and 40μM respectively. Our results indicate that combination dose significantly promote apoptosis and decrease cell proliferation. In addition, the level of mRNA Expression of MEK1,MEK2,ERK,Cyclin D1,NF-κB was decreas and PTEN was increase. Conclusion: In conclusion, we observed that SOR and CUR could be inhibits HCC proliferation and enhance anticancer effects.
