Abstract:

Background and Aim: Glycyrrhiza glabra L. a small perennial shrub native to the Mediterranean region, central and southwest of Asia. It is widely used in industry and medicine worldwide. The purpose of the present study was to callus induction and somatic embryogenesis induction of mature tissue in the shortest time.Methods: The experiment was conducted as a factorial experiment in a completely randomized design with four replications. The studied factors included two explants (leaf and petiole), and four murashige and skoog (MS) medium with growth regulator compounds.Results: The results showed that the highest callus induction (۸۰%) was observed in MS + ۰.۷۵ mg/l ۲.۴-D + ۱ mg/l BAP medium of leaf explants. The calluses obtained in MS + ۰.۷۵ mg/l ۲.۴-D + ۱ mg/l BAP medium had transparent color and low burn. Whereas in medium MS + ۱.۵ mg/l ۲.۴-D + ۱ mg/l BAP most calluses contained burns, chlorosis and high necrosis. The somatic embryos were increased (۶۰%) with the growth regulator ۲.۴-D (۱mg/l) increasing While ۲.۴-D concentration increased to ۱.۵ mg/l the percentage of somatic embryos decreased (۲۰%). The present study showed that embryos formed in MS + ۰.۷۵ mg / l + ۱ mg / l medium had the highest regeneration. Seedlings obtained from MS + ۰.۷۵ mg/l ۲.۴-D + ۱ mg/l BAP medium were intact, with thick, succulent stems and containing a huge number of leaves.Conclusion: Commonly, the highest callus induction percentage, somatic embryo, and regeneration were observed in MS ۰.۷۵ mg / l ۲.۴-D ۱ mg / l BAP medium. This medium would be advised for the next step such as genetic engineering and synthetic embryo production.
