Abstract:
Alzheimer's disease is classified as an advanced disorder of brain nerve cell destruction. The onset of thisdisease is due to the existence of two types of abnormal structures called ۱. Neurofibrillary tangles ۲.Nerve plaques. Aβ is the major constituent of amyloid plaques found in the brains of Alzheimer's patients.There are two common Aβ isoforms: the common form Aβ۱-۴۰ and the less common but amyloidogenicform of Aβ۱-۴۲ peptide. Crocin is a carotenoid of saffron flower stigma that has many medicinalproperties including antioxidant effect. The aim of this study was to evaluate the effect of crocinmicroencapsulation and to evaluate the temperature and pH stability of crocin microcapsules. Thermalstability and pH of crocin were determined at different temperatures (۶۰, ۷۰, ۸۰ and ۹۰ ° C) and pH levels(۲, ۴.۵ and ۶). Crocin decomposition showed first-order kinetics and the degradation rate constant (k)was significantly affected by pH and temperature (P <۰.۰۵). A high temperature and low pH showedhigher degradation (k) in crocin content. Chitosan-alginate nanoparticles were considered as a newvehicle for crocin to improve bioavailability properties. The in vitro release profile of crocin fromcomposite nanoparticles was investigated under simulated physiological conditions during incubationtime. Experimental reports collected in this study suggest that crocin-loaded nanoparticles could bepotential candidates for future treatments for Alzheimer's disease.
