Abstract:
Background: In infertility clinics, preserving high-quality spermatozoa for a long time is a necessity. Pentoxifylline (PT) and L-carnitine (LC) are two effective antioxidants in improving sperm quality. The present study aimed to evaluate the effects of Pentoxifylline (PT) and L-carnitine (LC on the quality of sperm DNA, and protamine content on prolonge storage for۱۲ days at ۴-۶°C.Materials and methods: The present experimental work included ۲۶ samples of normozoospermic men. After swim up, samples were divided into three aliquots as untreated control, LC-treated, and PT-treated groups and incubated for up to ۱۲ days at ۴-۶ °C. Thereafter, DNA denaturation and chromatin maturity were assessed on ۰, ۱, ۲, ۷, and ۱۲ days. Data were analyzed using a one-way analysis of variance.Results: The obtained data revealed that during the ۱۲-day storage, the percentage of spermatozoa with double stranded DNA and the percentage of spermatozoa with a normal protamine content was nearly unchanged in the three groups (p-value >۰.۰۵).Conclusion: Sperm DNA was resistant against changes during ۱۲ days at ۴-۶ °C in all groups
