Application of Molecularly Imprinted Polymer for SolidPhase Extraction and Preconcentration of Resorvastatin inbiological Sample Analysis abstract
A molecularly imprinted polymer (MIP) was synthesized and evaluated to selectivelyextraction of Resorvastatin, a HMG-COA reductase inhibitors in human urine samples. Thesynthesis of the MIP was performed in methacrylic acid (MAA) as the functional monomer,ethylene glycol dimethacrylate (EGDMA) as the cross-linker monomer, chloroform as thesolvent and Resorvastatin, template as the target drug. Ultraviolet spectroscopy, FT-IRspectroscopy, UV-VIS spectrophotometry, thermogravimetric analysis were performed tointerpret the specific recognition ability, the binding process and the characteristics of MIPs.In this study, solid phase extraction using the imprinted polymer has been optimized with theexperimental design approach for Resorvastatin, with regard to the critical factors whichinfluence the molecular imprinted solid phase extraction efficiency such as sample pH,volume of elution solvent, washing solvent and sorbent mass. Batch rebinding capacity ofResorvastatin was determined from the derived Freundlich isotherm and was found to be1.45μmolg-1. The recovery and limit of detection for urine sample were 98% and 5.9 μgL−1,respectively. The results showed that, compared with other reports, MIPs synthetized in thisstudy showed high adsorption capacity besides specific recognition ability. The synthesizedMIP has enabled the direct percolation of human urine and the easy elimination of endogenouscompounds from it with simple aqueous washing of the MIP-SPE packing. HPLC analysishas confirmed the high extraction recovery of resorvastatin, from such urine samples with useof proposed MIP-SPE system.
