The relationship between IL-۱۰ (-۸۱۹ C/T) polymorphism and Hepatitis B infection

Background and Aim: Anti-inflammatory cytokine IL-۱۰ is important and single nucleotide polymorphisms in the interleukin-۱۰ gene promoter is associated with HBV infection. Especially single nucleotide polymorphisms in the promoter region of the gene IL۱۰, at position -۱۰۸۲, -۸۱۹, -۵۹۲ is associated with production of IL۱۰. Therefore, the present study was to determine the effect of IL-۱۰ -۸۱۹ polymorphism with chronic HBV infection in Iranian patients. Materials and Methods: A total of ۲۰۰ subjects (۱۰۰ chronically infected hepatitis B patients and ۱۰۰ blood donors) with an age range of ۱۸-۵۹ years were included in this study randomly.The study in ۱۳۹۲ on samples of patients referred to clinical laboratory blood transfusion was performed. Genomic DNA was extracted from Buffy coat layer with using of salting out procedure.Allele Specific Polymerase Chain Reaction Method was used for determining polymorphisms -۸۱۹ (C/T) in the IL-۱۰ gene promoter.Electrophoresis was performed on a arose gel ۱.۵ per cent for the detection of PCR products. Data were analyzed using Chi Square analysis Results: The frequency of Homozygote genotype IL-۱۰ -۸۱۹ (TT, TC, CC) was in patients with HBV ۸۴%,۱۲% and ۴% and ۸۹%,۱۰% and ۱% in healthy individuals respectively  and no significant difference was observed between the two groups in terms of genotype (P =۰.۸). Variant of C allele frequency was in patients with HBV ۱۰% and ۶% in healthy individuals. There was no statistically significant difference between the two groups (P=.۰.۶). Conclusions: Our results showed that there was no correlation between polymorphism IL-۱۰-۸۱۹(T/C) in control group and chronic Hepatitis B infection.
