Preparation and evaluation of hyaloronic acid /Chitosan-coated PCEC polymeric nanoparticles for targeted co-delivery of paclitaxel and chrysin to prostate cancer cells abstract

In recent years, extensive efforts have been made to overcome the limitations of chemotherapy. The objective of the present study was to prepare and evaluate the targeted polymeric nanoparticles for co-delivery of anticancer drugs to PC3 cells to improve PTX therapeutic efficacy and reduce its side effects. In this study, the prepared nanoparticles were analyzed for their chemical structure, particle size, zeta potential, surface morphology, drugs encapsulation efficiency, loading capacity, and drugs release profile. The antitumor effect were evaluated in PC3 and HUVEC cell lines using the MTT assay. The results exhibited that NPs have a homogeneous spherical structure and mean particle size and zeta potential value were 219 nm and -11.6 mv, respectively. The encapsulation efficiency was 78.6% and 93.28% for CHR and PTX, respectively. The drug release profile showed that the release of both PTX and CHR in acidic conditions was significantly higher than the physiological conditions, which could be due to the pH-sensitive coating. The results demonstrated that the dual drug-loaded NPs have a significant effect on reducing the survival of PC3 cancer cells compared to single drug-loaded NPs or free dual drugs. Our findings indicated that targeted PCEC nanoparticles loaded with PTX/CHR can be considered as a suitable candidate for the effective treatment of the prostate cancer.
