Abstract:
Background: Cancer is the second leading cause of deathworldwide. Chemotherapeutic drugs are only partially effectivedue to low bioavailability and susceptibility to resistanceacquired by cancer cells leading to poor treatment Results: Inaddition, due to differences in pharmacokinetics and pharmacodynamicsproperties, these agents often cause adverse effectsand might lead to drug resistance. This study aims to investigatecurcumin as a therapeutic agent for multi-drug resistance(MDR) in cancer cells.Materials and Methods: PubMed, Scopus and Web of Sciencewere searched up to April ۲۰۲۳. Most of the evidence is in-vivoand in-vitro studies that demonstrated the mechanism and efficacyof curcumin on chemo-resistant cancer cells.Results: It has been suggested that MDR in cancer cells is dueto the upregulation of ABCC۲, P-glycoprotein (P-gp), and thedownregulation of DCK, TK۱, and TK۲. Curcumin decreasedthe expression of ABCC۲ and increased the expression of DCK,TK۱, and TK۲. In addition, curcumin suppressed P-gp expressionvia the inhibition of the PI۳K/Akt/NF-kappa B pathway,which had a key role in MDR. Also, Curcumin modulatesmultiple molecular targets such as STAT۳, EGFR, FOXO۳a,TGF-β, eIF۲α, COX-۲, Bcl-۲, PI۳KAkt/mTOR, ROS, Fas/FasL, Cdc۴۲, E-cadherin, MMPs, and adiponectin in MDRcancer cells.Conclusion: Curcumin could reverse the effect of MDR andmaximize the clinical efficacy of anticancer drugs in resistantcells while minimizing their side effects in future cancer treatment.However, more research needs to confirm or refute this
