Deciphering the Impact of the L858R Mutation in EGFR: Computational Insights and Therapeutic Implications in Non-Small-Cell Lung Cancer abstract

This study delves into the L858R mutation in the Epidermal Growth Factor Receptor (EGFR) and its impact on lung cancer, specifically non-small-cell lung cancer (NSCLC). Employing computational tools like the DynaMut web server and PSIPRED, we assessed evolutionary conservation, protein stability, flexibility, secondary structure, and interatomic interactions of the EGFR protein. Our findings suggest that the L858R mutation, while highly conserved, induces significant changes in the protein's stability, flexibility, and secondary structure, potentially influencing its function. These alterations offer insights into the development of targeted therapies for NSCLC, underscoring the mutation's impact on protein behavior and therapeutic implications. The study highlights the importance of computational predictions in understanding the molecular basis of cancer and suggests the need for experimental validation in future research.
