Chemo-Phototherapy of Breast Cancer Cells by In situ GrownPolydopamine Nanoparticles in Alginate Hydrogels abstract

Breast cancer is one of the most common cancers in the world. Surgery is the preferred treatment forthis cancer, but the risk of local recurrence and metastasis of the remaining cells after surgery is still asignificant concern. Here, we prepared a hydrogel of alginate-dopamine (Alg-DA) and polydopaminenanoparticles (PDA NPs), which exhibited excellent photothermal capacity after NIR irradiation (808nm, 1 W/cm2). The PDA NPs were used at different concentrations to investigate their effect on celladhesion and photothermal properties. The hydrogels showed acceptable biocompatibility and celladhesion after seeding MDA-MB-231 cells and provided a suitable environment for cell growth. Inthe subsequent phase, we refined our model by culturing MDA-MB-231 tumor cells with humanprimary fibroblast cells to create a model that more closely resembles the in vivo environment andbetter mimics the in vivo situation. The chemo-photothermal (CPT) ability of the hydrogels wasinvestigated by the simultaneous use of docetaxel (DTX) and laser irradiation. The results indicated asynergistic effect of the drug and the laser on the death of cancer cells in an in vitro environment. Thistherapeutic method can potentially prevent the spread of tumor cells and metastasis and can be usedafter surgery to eliminate residual cancer cells.
