Mitochondria-Targeted Antioxidants (MitoQ & Mito-TEMPO) Improve Motility and Mitochondrial Activity of Cooled Rooster Sperm

This study compared the effects of mitochondria-targeted antioxidants (MitoQ and Mito-TEMPO) versus untargeted antioxidants (SOD and CoQ۱۰) on rooster sperm motility and mitochondrial activity during cooling (۴°C for ۴۸ hours). Semen samples were collected from Ross-۳۰۸ roosters and divided into eight groups: control, SOD (۱۰۰ U/mL), CoQ۱۰ (۱ _x۰۰۰۳_۹ M), MitoQ (۵۰ and ۱۰۰ nM), Mito-TEMPO (۵۰ and ۱۰۰ _x۰۰۰۳_BCM), and MitoQ+Mito-TEMPO combination. Total motility (TM), progressive motility (PM), and mitochondrial activity potential (MAP) were assessed at ۰, ۲۴, and ۴۸ hours. At ۲۴ and ۴۸ hours, mitochondria-targeted groups showed significantly higher TM, PM, and MAP compared to control, SOD, and CoQ۱۰ groups (P<۰.۰۵). The combination group (MQ/MT) exhibited the best results. Mitochondria-targeted antioxidants, especially their combination, are recommended for rooster semen cooling.
