Abstract:

We have reported the fabrication of Herceptin functionalized liposomes containing PVP-stabilized goldnanoshells as a promising hybrid structure for induction of apoptosis in HER2 overexpressing breast cancer cell. Sizeand optical properties of gold nanoshells were evaluated using TEM and UV-Vis spectroscopy. The effect offunctionalization on cellular uptake of nanoparticles (NPs) was assessed using two cell lines of BT-474 human breastcarcinoma and Calu-6 human lung carcinoma. Uptake of NP-loaded immunoliposomes by BT-474 cells was about 5and 2 folds than that of NPs and NP-loaded liposomes, respectively. Also, rising temperature of the suspension of goldnanoshell during irradiation by continuous wave Ar laser was recorded using digital thermometer. Finally, applicationof immunoliposome containing gold nanoshells for NP-mediated laser hyperthermia studied in vitro by terminaldeoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay which revealed 44.66% apoptosis afterillumination.
