Correlation between various k-ras gene codons (codon12 and codon 13) mutations with ovarian cancer abstract

Background: k-ras protein is a GTPase and is an early player in many signaling transduction pathways. Multiple studies have shown that patients with tumors harboring mutation in k-ras are unlikely to benefit from anti EGFR antibody therapy. This study aimed to compare the prevalence of k-ras gene (codon 12 and codon 13) mutations in patients with ovarian cancer Method: The DNA was extracted from paraffin embedded tissues and subjected to k-ras mutation analysis by nested PCR and RFLP analysis. Result: In our study none of 30 patients had mutations in codon 13 and 3 of them had mutation in codon 12 of k-ras gene. Conclusion: The results of this study revealed that mutation at k-ras codon 12 is correlated with mucinous type of ovarian cancer but no correlation was found between codon13 and ovarian cancer.
