Study of the Anticancer Effects of Methotrexate in the Presence of UV Light in MCF-7 Cell Line abstract

Cancer is one of the most wide-spread diseases. breast cancer is the second most commoncancer among women. Today, the most common treatments against cancer is radiotherapyand chemotherapy, but these approaches are non-specific mechanism. Therefore, it isconsidered that using of targeted therapies that specifically kills cancer cells and lessdamage for normal cells. Photodynamic therapy is one of these methods. Photodynamictherapy consists of three main components photosensitizer, light and oxygen. None of thesesubstances are not toxic alone, but together they have initiated a photochemical reaction thatproduced reactive oxygen species.In this study, methotrexate use as photosensitizer and anticancereffects of it in the presence of light evaluated on breast cancer cell line. MCF-7 cellline cultured in RPMI-1640 medium then treated with methotrexate, after 8 hours, cellsirradiated with UV light for 40 second. then incubated for 48 hours and cytotoxicity effectmeasured MTT assay. Concentration of methotrexate that in presence of light killed 50%growing cell calculated by MTT, LC50 concentration was 30ÂμM.LC50 of the methotrexate inpresence of UV light was 30ÂμM, this concentration of methotrexate and this dose of UV lightwas safe for the cell. simultaneous treatment showed that methotrexate act as suitablephotosensitizer. Using methotrexate as a photosensitizer could cause more cytotoxic effects,therefore LC50 concentration get lower and finally side effect of methotrexate could bedecreasing.
