Abstract:
Methylenetetrahydrofolate reductase (MTHFR) is the rate-limiting enzyme in the methyl cycle. MTHFR encoded by the highly polymorphic MTHFR gene that in human has been located at 1p36.3chromosomal region. It does comprise eleven exons and plays a key role in homocysteine plasma level which is associated with the risk of breast cancer. The single nucleotide polymorphism (SNP)rs1801133 also known as the C677T, Ala222Val or A222V is thought to be associated with the predisposition of breast cancer and subsequent immune response in the different populations. The homozygous re1801133 (T; T) nearly has the 30% MTHFR enzyme activity while the heterozygotes re1801133 (C;T) nearly has the 65% activity in compare with the most prevalent re1801133 (C;C)genotype. The aim of present study is the evaluation of rs1801133 in MTHFR with the risk of breastcancer in patients with carcinoma of breast and controls. Our study conducted on 140 patients with breast carcinoma and equal controls. The TaqMan real time PCR technique was applied for genotyping of participants. The correlation of both variants and demographic data were investigated with the riskof breast cancer. Our obtained results showed that the allele T and TT genotype had the higher prevalent in patients (p value=0.0001) and (p value=0.003) than control group. Indeed, the correlationsof menarche and underlying hormonal disorder with the risk of breast cancer were evaluated.The current study clarified the relationship of MTHFR rs1801133 with the risk of breast cancer in Iranianfemales. The mutation in MTHFR gene rs1801133 could be used as the predictive biomarker for breast cancer. Keywords: breast cancer, MTHFR gene, allele, polymorphism, genotyping
