Abstract:
Objectives and background: development of nanomaterials starts to catch medical scientist attentions into amazing physiochemical properties of particles in Nanoworld. Cerium oxide nanoparticlescomposed of a core element of lanthanide’s series of the periodic table, cerium, surrounded with a close mesh of oxygen and have strong antioxidant activity. The purpose of this study is to investigateanti-oxidant properties and cytotoxicity effects of cerium oxide nanoparticles synthesized via green chemistry on colon cancer and normal cell lines. Materials and Methods: MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) cell viability assay was used in order to compare cytotoxicity of cerium oxide nanoparticle between HT-29 colon cancer and HUVEC normal cell lines. Cells wereseeded in 96-well plate and after 24 h-incubation, different concentrations of Cerium oxide nanoparticles (0, 62.5, 125, 250 and 500 μg/ml) were used to treat cells. Treatments performed at 24 hour time interval. Then treatments removed and MTT was added to plates and after dissolving dye with DMSO, Plates were put in ELISA reader and optical absorbance was read at 570 nm. Results: results showed that cerium oxide nanoparticles synthesized in Rubia tinctorum extract, are able to inhibit colon cancer cells and value of IC50 for HT-29 cell lines were 260 μg/ml, while the IC50 value for HUVEC cell line was more than 500 μg/ml. In addition, results showed that the inhibitory effects of cerium oxide nanoparticles on colon cancer cell line are more than normal colon cell lines. Conclusion: our findings indicated that cerium oxide nanoparticles synthesized through green chemistry using Rubia tinctorum extract have potential anti-cancer properties and more studies in order to confirm this property are recommended.
