Abstract:

Introduction and Aim: Cancer cases is on the rise around the world. Breast cancer is the most common, among women. The most ideal anti-cancer drug is a combination that can without harming healthy cells by targeting and destroying cancer cells demonstrate its therapeutic effects. In this study we aimed to formulate the effect of Transfrozom on cancer cell line 4T1.Methods: First, phosphate buffer and pure Hydroxyurea were prepared. Then, Standard curve of Hydroxyurea was drown and pure phosphatidylcholine was prepared. For Making Transfrozom particle diameter and surface charge were measured by Zeta sizer. Cryo solution was used for freezing which prepared of 90 ml FBS and 10 ml DMSO. After defreeze frozen cells, MTT method was used for cytotoxicity diagnosis. Also, IC50 of the formulation obtained, by PHARM software. Results: The average size of control Transfrozomal nanoparticles without drug and the surface charge were reported 78nm and -10.03, respectively. Also, the average size of Nano-Transfrozomal particles containing Hydroxyurea and the surface charge were 52nm and -8.07, respectively. The amount of absorption of the samples in successive hours, also Hydroxyurea concentrations in the samples were assessed. The survival rate of cells determined by MTT test.Conclusion: In this research, manufacturing methods were tested Transfrozom, and measuring the characteristics such as size, surface charge, the drug trapping efficiency, the amount of drug release, appropriate proportion of the lipid composition and surfactants, also optimal method of making nanoparticles were obtained. Finally, formulation effect on cancer cell line 4T1, was compared with free drug.
