Optimization of Plant Peroxidase Purification abstract
Partial purification for plant peroxidase from white radish was optimized. Precipitation by ammonium sulfate and aqueous two phase system (as two parallel methods) were used. Polyethylene glycol/ammonium sulphate/NaCl (27:6.0:2.0%, w/v) system followed by gel filtration chromatography gave purification factor of about 34 fold. On the other hand, ammonium sulfate precipitation (40-80% degree) followed by gel filtration gave only 16.5 purification fold. Hence, aqueous two phase system was more efficient and useful primary purification step since it was less laborious, less time consuming, and led to more purification factor. The partially purified white radish enzyme was stable at pH range 4.0-8.0 and had an optimum pH at 4.0. The enzyme was stable at 65°C with an optimum activity at 55°C.
