Abstract:
IntroductionNowadays, diabetes afflicts more than 300 million people in the worldwide. It has been proven that type I diabetes is an immunologically-mediated devastation of the insulin-producing cells (IPCs) in the pancreatic islet. It seems that stem cells that can differentiate to β- cells may be a new promising therapeutic application. Pancreatic and duodenal homeobox factor 1 (PDX1) is a transcriptional factor necessary for the pancreas development. PDX1 plays a distinct role in proliferation and differentiation.MethodsIn this study, we have used a lentiviral vector to deliver the PDX1 gene into MSCs to generate IPC. After transduction, we measured insulin and C-peptide production in these cells .ResultsInsulin and PDX1 gene expressions were evaluated by real-time polymerase chain reaction (RT-PCR) and immune cytochemical analyses.ConclusionWe proved this approach had a suitable efficacy for treatment of diabetes
