Abstract:
Acute and chronic wounds affect millions of people worldwide as well as burns and they are the major causes of full thickness skin wounds. In this study, the process of skin wound healing was investigated by examining the effect of activated Chitosan-Gelatin polymeric scaffold with Bioglass Nanoparticles containing silver in a mouse model. Morphology, Biocompatibility and Bioactivity of the Chitosan-Gelatin composite scaffolds with Bioglass particles containing silver were investigated. A total of 24 mice were divided into two treatment (scaffold) and control groups. A deep circle-shape wound, with a diameter of 5 mm, was created on the mouse skin using a biopsy punch. Wound healing of both control and treatment groups was examined and compared clinically and histopathologically on days 7, 14, and 21 post operation. The healing process was investigated using H&E staining and a specific trichrome massone staining against CD31 and CD34 markers to study angiogenesis. Our results represented the composite scaffolds containing Bioglass silver accelerated wound contraction and healing compared to control groups. The Chitosan-Gelatin composite scaffolds with bioglass containing silver can be considered as an appropriate scaffold for skin tissue engineering applications
