Green synthesis of Ag nanoparticle with Sambucus ebulus rhizome. Extract and evaluation of antibacterial activity

Abstract:

Background :As microbial resistance against antibiotics is increasingly growing, it is necessary to develop new remedies. Silver nanoparticles have shown significant antibacterial properties. Synthesis of silver nanoparticles mediated by plant extracts could be considered as an alternative in treatment of various infections. Method: In this study silver nanoparticles have been synthesized using Sambucus ebulus rhizome extract. Optimum temperature, concentration and also the best pH for production of silver nanoparticles were found. Formation of the particles has been investigated by UV-vis spectra, EDS, XRD and SEM techniques.The antibacterial effect of these particles then has been studied. Ciprofloxacin was used as positive control. Determination of MIC and MBC were carried out. Result: Optimum condition for synthesis of silver nanoparticles was at 65˚C, pH=10 and 30 minutes after the addition of silver nitrate to Sambucus ebulus rhizome extract. The particles remained stable until two months after synthesis. The approximate size of silver nanoparticles was 30 to 40 nanometer. The best MIC and MBC was observed for Pseudomonas aeruginosa 4 and 30 mg/ml. Conclusion: Plant natural products are involved in the reduction to synthesize nanoparticles. They are particularly participating in defence mechanisms to produce several chemical compounds like polyphenols.This study revealed that synthesized nanosilver by Sambucus ebulus rhizome extract shows strong antibacterial effects on Pseudomonas aeruginosa, and moderate antibacterial activity on Staphylococcus aureus and Escherichia coli.
