THE EFFECT ESSENTIAL OIL OF THE NIGELLA SATIVA ON NICOTINE INDUCED CELL DEATH IN PC12

Abstract:
Background and Aim : High concentration of nicotine induces cell death in central nervous system cells which can affect the health of nicotine-exposed people. Some of the Nigella Sativa s therapeutic properties have been investigated but its protective power against neuron cell death not studied yet. Therefore, we studied the protective effect of Nigella Sativa oil on nicotine-treated PC12 cells.Methods : The cell viability measured by trypan blue assessment. The cell proliferation was calculated by MTT test. LDH test was performed to investigation of cell cytotoxicity. TUNEL test was used to show apoptosis. NO production was calculated through Griess reaction. The Rat inflammatory cytokine assay kit was used to measurement of inflammatory cytokines concentrations. Caspase-3 activity was assessed by the caspase activity colorimetric assay kit and mitochondrial membrane potential was studied by Rhodamine123 florescence dye.Results : 100 mM of nicotine reduced cell viability, proliferation, mitochondrial membrane potential, and increased cell cytotoxicity, NO production, inflammatory cytokines concentrations, caspase-3 activity, and DNA fragmentation significantly. N. sativa-treated cell cutlers increased cell viability, proliferation, mitochondrial membrane potential, and decreased cell cytotoxicity, NO production, inflammatory cytokines concentrations, caspase-3 activity, and DNA fragmentation which caused by apoptotic concentration of nicotine in PC12 cells.Conclusion : The main finding of our study was that N. sativa oil suppresses the nicotine-induced cell death in PC12 neuron-like cells. It inactivates the apoptosis process through inhibition of the NO production, and inflammatory cytokines synthesis.
