Abstract:

The explants were excised from mature, 12 years old, selected within plus trees, in Fars and Khuzestan provinces (Iran). Various Explants were compared, to choose the best kind of micro cutting. All other experiments were done to find out the most suitable methodology of sterilization Using Benomyl 20 g/l, Ethanol 70% for 60 seconds, Hypocloride 20% for 15 minutes. The mono and binodal microcuttings were cultured in MS medium containing BA 0.5 mg/l in estabilishment phase. Within numerous hormonal treatments for proliferation stage the best one resulted in: BA 1.5 mg/l and IBA 0.1 mg/l. Agglomerted and very condensed shoots were grown. They were excised in small pieces and were transfered to a new medium containing GA3 0.5 mg/l + BA 0.1 mg/l. Contemporanously, was applied a period of 10 days dark treatment. At the end of this period we obtained shoots, 6 cm height, and toghether with functional long roots. For acclimation was used a proportional mixture of perlyte: 1, sand: 2, and manure 1. This phase was carried out successfully and acclimated plantlets, 10 cm height, were replanted into the bigger pots. Micropropagated cloned plantlets, 42cm height, were transplanted in the field. After 6 months from transplanting, the cloned trees, 415 cm height, were highly adapted demonstrating 0% of mortality during adaptation test.
