Micropropagation of Aglaonema abstract
Aglaonema is one of the most important in door plants in the word. The plant material used in this experiment is cultivar Chinese evergreen (Aglaonema "Siam Aurora") with Piebald red and green leaves which were kept in the ornamental greenhouse and irrigated with fertilizer 18-18-18, regularly. Bed used for Aglaonema is Coco peat and perlite (1:1). In this study lateral bud stem and leaf explants are used. Basal medium for callus experiment were used Murashig and Skoog (MS) supplemented with TDZ at 3 levels (0.5, 1 and 2 mg.l-1), NAA at 2 lavels (0 and 0.2 mg.l-1), 2,4-D at 5 levels (0, 0.5, 1, 2, 3 mg.l-1) and Kin 0.33 mg.l-1.. Lateral bud explants were used for direct Regeneration experiment in a basal medium Murashig and Skoog (MS) supplemented with TDZ at 2 levels (1 and 1.5 mg.l-1), BAP at 3 lavels (3, 6 and 9 mg.l-1), NAA at 1 level (0.05 mg.l-1). Leaf explants in 0.5 mg.l-1 2,4-D and 2 mg.l-1 of TDZ treatment showed the best results for volume callusing and production embryogenic callus. In lateral bud explants, 1.5 mg.l-1 TDZ showed the best results. Basal medium 1/2 MS supplemented with 2 mg.l-1 IBA has most number and length of roots in rooting of in vitro explants.Key word: Aglaonema, Micro propagation , Auxiliary bud ,leaf ,Rooting
